Recruiting a character is the only thing that counts as playing the character card.

Boost used to refer specifically to recruitment. But when the keyword was introduced to other character types, the definition was adjusted from "recruit" to "play." This worked because the two wordings were equivalent for characters.

Using Substitute, or for that matter any other power on a card, is playing an effect, but it's not playing the card. As a result, using a payment power will never count for or be prevented by the Joker AE.
